Analyst Report: MRNA
1. EXECUTIVE SUMMARY
Moderna fell 17.57% to $202.63 on Thursday, October 22, on volume of 49,777,400 shares, 2.1x its 50-session average, after Phase 3 data for its melanoma cancer vaccine (intismeran autogene plus Keytruda) leaked on social media ahead of the planned ESMO presentation. The combination cut the relative risk of recurrence or death by 42% against Keytruda alone, according to BioPharma Dive, which sits below the 44% seen in the smaller Phase 2 trial and at the low end of the 40–50% range analysts had expected. Analysts quoted by BioPharma Dive and Stocktwits mostly called the reaction a sell-the-news event after a run that took the stock to a 52-week high of $254.48 on October 21. Biotech peers rose over the same period, so the decline belongs to Moderna alone. The full presentation on October 24 and third-quarter earnings on November 6 are the next tests.
2. THE CATALYST
Wire headlines began at about 18:10 UTC on October 22, after screenshots of the data circulated on social media before the companies' planned release (Forbes, BioPharma Dive and AJMC, all October 22). AJMC and BioPharma Dive differ slightly on the exact timing of the leak, and both place it on that Thursday. ESMO then released the full study details early.
| Item | Figure | Source |
|---|---|---|
| Relative risk of recurrence or death vs Keytruda alone | -42% (median follow-up 24.6 months) | BioPharma Dive |
| Distant metastasis or death vs Keytruda alone | -38% | Stocktwits |
| Phase 2 trial result (2022) | -44% (HR 0.56) | Moderna 8-K, December 2022 |
| Five-year Phase 2b result | -49% | Drugs.com, June 1, 2026 |
| Analyst expectation range | 40–50% | BioPharma Dive |
| Level Moderna had called clinically significant | 35% | BioPharma Dive |
Overall-survival data are immature but trend favourably, and no new safety signals were reported (Stocktwits). AJMC noted that investors had anchored on the earlier KEYNOTE-942 result (HR 0.51, per RBC), which left the primary endpoint at the low end of expectations.
The stock closed at $245.81 on October 21 and opened at $240.12 on October 22. It touched $180.80 intraday and closed at $202.63. Outlets cited intraday declines between about 18% and 25%, which reflect the timing of each report. The close of -17.57% is the figure that applies.
3. COMPANY PROFILE
Moderna is a messenger RNA biotechnology company in the Healthcare sector. Its portfolio spans respiratory vaccines (Spikevax, mNEXSPIKE, mRESVIA), latent virus vaccines, norovirus, public health vaccines and oncology therapeutics, including intismeran autogene. Merck co-owns the Keytruda combination programme. Other cancer-vaccine developers include BioNTech, which Mizuho's Jared Holz named as being on his radar (Stocktwits).
Revenue fell to $1.9bn in 2025 from $3.2bn in 2024, and the company is seeking growth beyond COVID vaccines, with a target of cash breakeven around 2028 (Stocktwits). Earlier in October, Moderna and Merck expanded their collaboration with Tempus to support possible commercialisation of the cancer vaccine, and Moderna joined the Nasdaq-100 (Stocktwits). The market capitalisation is $80.90bn and the forward P/E is -44.7, which reflects expected losses. The August 19 topline announcement had said only that the combination met its primary endpoint at an early pre-planned check (BioPharma Dive).
4. DEEP DIVE ANALYSIS
Do fundamentals justify the move? The data show a 42% reduction in recurrence or death, a result above the 35% level Moderna had called clinically significant. TD Cowen's Tyler Van Buren called the findings compelling and in the exceptional range, and Bernstein said the data validated the platform (Stocktwits). The size of the fall comes from positioning: the stock had risen from $54.49 on July 29 to $245.81 on October 21, a gain of roughly 351% over those 60 sessions, with the largest single-day percentage gain on record on August 19 (Stocktwits). Even after the fall, the close of $202.63 is 271.87% above the close of 60 sessions ago. Analysts mostly called the move sell-the-news, including William Blair's Myles Minter (BioPharma Dive). Mizuho's Holz said the data "weren't bad" but the bar was too high.
Peers and read-across. Merck fell about 1% intraday (BioPharma Dive), much less than Moderna. The biotech peers in the panel rose over the period: ABBV +0.14%, AMGN +2.04%, GILD +2.36%, VRTX +2.03%, REGN +1.67% and BIIB +5.34%. The S&P 500 fell 1.21%. Holz said Merck looked more compelling for investors bullish on the programme (Stocktwits).

| Firm | View | Source |
|---|---|---|
| TD Cowen | Hold, target $90 (raised from $40 on October 21) | Investing.com, MarketBeat, Stocktwits |
| RBC (Trung Huynh) | "Materially less impressive" than the earlier trial; "approval is likely, commercial success far less certain" | BioPharma Dive, Stocktwits |
| BMO | Results not enough to build confidence; efficacy beyond melanoma unproven | Stocktwits |
| Wells Fargo | Merck concerns may be overdone; recurrence-free survival beat expectations | Stocktwits |
| Bernstein | Market Perform, $45 target (single source, unconfirmed) | Stocktwits |
MarketBeat's consensus is Hold with a $103.95 average target and 7 Buy, 14 Hold and 4 Sell ratings. That average target sits well below the $202.63 close, so most published targets have not caught up with the share price. TD Cowen raised its peak intismeran revenue estimate to about $3.4bn by 2035 from $830m.
Bull case. A 42% reduction with no new safety signals supports likely approval, in RBC's view. The platform has now produced positive Phase 2 and Phase 3 data, and the commercial partnership with Merck and Tempus is in place. Holz noted that the market still lacks evidence in renal cell and lung cancer, so further readouts offer room for upside.
Bear case. The Phase 3 result is below the 44% of the Phase 2 trial and the 49% five-year figure. RBC doubts commercial success, and BMO flags unproven efficacy beyond melanoma. Revenue is shrinking, and the valuation of $80.90bn leans on a programme whose revenue TD Cowen puts at about $3.4bn at peak. In my inference, the gap between the consensus target of $103.95 and the price leaves room for further declines if the October 24 presentation adds no new positives.
5. TECHNICAL SNAPSHOT
The close of $202.63 is above the 50-day average of $168.42 and the 200-day average of $81.34, and sits 20.4% below the 52-week high of $254.48 set a day before the drop. The nearest support is the close of $194.82 reported 20 sessions ago on September 24, while the closes in the recent table offer $213.74 (October 12) as the nearest resistance, with $218.01 (October 16) above it. Volume of 49,777,400 was 2.1x the 50-session average and well above any other session in the table, the closest being 28,947,300 on October 9. That shows heavy distribution on the leak.
6. RISK FACTORS
- Further data detail on October 24 at the ESMO Presidential Symposium (LBA1), with simultaneous New England Journal of Medicine publication, may revise views on distant metastasis and overall survival (Merck, CancerNetwork).
- Pancreatic (Abstract 3151P) and early-stage NSCLC (2550TiP) posters follow on October 25–26 (Moderna).
- Third-quarter earnings on November 6 carry a consensus EPS estimate of -1.3 and a revenue estimate of $892.38m.
- Efficacy outside melanoma is unproven (BMO, Holz), and commercial uptake remains uncertain (RBC).
- The stock's 556.61% year-to-date gain leaves a large cushion of profit for holders to realise.
- The Bernstein $45 target comes from a single source and is unconfirmed.
7. ACTIONABLE OUTLOOK
Short term (1-2 weeks): The October 24 presentation and journal publication supply the next information, with distant-metastasis and overall-survival detail the focus. Support at $194.82 and the intraday low of $180.80 mark the range where buyers stepped in. A recovery toward $213.74 would need supportive detail from the presentation.
Medium term (1-3 months): The November 6 earnings report and any regulatory filing news drive direction. Analysts' targets average $103.95, so sentiment among published ratings remains cautious relative to the price.
Long-term thesis: The data confirm that the vaccine works in adjuvant melanoma, and the commercial case rests on expansion into other tumour types and on revenue growth beyond COVID. The 42% result left that thesis intact and reset the expectations priced into the shares. This is analysis only and not personal financial advice.
